James is sidelined for around four weeks and will miss the opening of the league campaign due to sciatic nerve pain, LA Lakers stated.
The player, aged 40, agreed to a new agreement with the LA team in June and is poised to be the first athlete to compete across 23 individual campaigns.
Sciatic pain involves pain in the sciatic nerve that travels down the spine and through the leg.
James has not taken part in preseason and was out for the games against the Golden State and the Phoenix Suns earlier this month as a result of the sciatic problem.
The veteran is projected to sit out for each of the five of the Lakers' games in the month of October, including the first game against Golden State Warriors.
LeBron James, the league's top scorer, has never previously missed the beginning of the campaign in his 22-year career.
His tally of 1,562 regular-season appearances is only 50 away than overtaking former Boston star Robert Parish's league record.
The title winner appeared in 70 out of the LA Lakers' 82 games the previous campaign and competed in 71 games in the previous year.
In the most recent campaign, the player posted 24.4 points, 8.2 assists per game and 7.8 rebounds per game per game and finished sixth in the Most Valuable Player vote.
Los Angeles Lakers qualified for the postseason in the 2024-25 season but fell four games to one against the Minnesota in the first round.
